In this bachelor thesis was described the reproductive system of a bull, hormonal regulation, sperm production and sperm, was also focused on the collection and sampling methods ejaculate. Work dealt with of studies of morphologically abnormal sperm and their influence on the fertility of bulls. At morphological defects been described primary and secondarily changed spermatozoa. Examination of native ejaculate was divided into microscopical, microbiological and special. At the end of this bachelor thesis was given the chapter on automated scoring systems ejaculates. Of those systems been described system for evaluating ejaculate CASA and flow cytometry.

Bachelor's thesis describes the qualitative and quantitative indicators of roosters ejaculate. Literary review deals with the reproductive system of domestic fowl, the composition of the ejaculate, sperm morphology and spermatogenesis. It also deals with macroscopic and microscopic evaluation of semen. It altso describes the morphological defects in sperm. The methodology to evaluate the qualitative and quantitative indicators of ejaculate of two laying breeds - Rhode Island White and Brindle Plymouth Rocsk. Both breeds have fewer than 20% of morphologically abnormal sperm. Most of the changes were represented at the connecting piece, the head and a tail.
Semen quality of stallions involved in breeding

The main objective of the study was to determine the seminal characteristics of warmblooded stallions, to assess a relationship between motility parameters of spermatozoa and sperm head dimensions (Experiment 1) and to compare the ability of three milk-based extenders to preserve the motility and viability of cooled shipped semen (Experiment 2) using Sperm Class Analyzer (SCA, MICROPTIC SL). In Experiment 1, 32 and 36 ejaculates were collected during breeding seasons 2016 and 2017 from 10 clinically healthy and fertile warmblooded stallions (age 3- 22 years). The volume of ejaculate (mL), sperm concentration (M/mL), motility of sperm with kinematic parameters, viability of sperm (%) and sperm morphometry with morphometric parameters were evaluated. In Experiment 2, 21 ejaculates from 11 stallions were collected before the beginning of the breeding season 2018 and diluted with Kenney, EquiPlus and INRA 96. Spermatozoa viability (VIT, %), total motility (MOT, %), progressive motility (PMOT, %) and selected kinematic parameters were evaluated for 63 semen samples. These parameters were determined 2 hours after semen processing and then in 24, 48 and 72-hour intervals of storage at 5 °C. The following average values were found in the obtained ejaculate samples for the observed periods of the breeding seasons 2016 and 2017 - volume of ejaculate 49.22 and 56.77 ml, sperm concentration 208.20 and 222.19 M/ml, MOT 82.44 and 81.38 %, PMOT 37.34 and 35.46 %, VIT 60.71 and 42.91 %, morphologically normal sperm 75.00 and 64.50 %, curvilinear velocity (VCL) 62.67 and 60.26 ?m/s, straight-line velocity (VSL) 32.10 and 27.55 ?m/s, average path velocity (VAP) 48.14 and 44.35 ?m/s, linearity coeficient (LIN) 50.64 and 44.89 %, straightness coeficient (STR) 66.56 and 62.01 %, wobble coeficient (WOB) 75.74 and 72.12 %, amplitude of lateral head displacement (ALH) 2.59 and 2.90 ?m, beat- cross frequency (BCF) 7.32 and 7.49 Hz, head length (L) 6.33 and 6,41 ?m, head maximum width (W) 3.01 and 3.11 ?m, head area (A) 15.65 and 16.37 ?m2, head perimeter (P) 16.26 and 16.55 ?m, elongation (Elong ) 0.36 and 0.35, ellipticity (Ellip) 2.11 and 2.07, roughness (Rugo) 0.74 and 0.75 and regularity (Regu) 0.96. There where significant differences among stallions in all evaluated parameters of sperm (P < 0.05). Furthermore, the influence of age and fertility of stallions was also demonstrated for the observed parameters. MOT, PMOT, VCL, VSL, VAP, LIN and WOB were positively correlated with area, width, length and perimeter of the head and negatively correlated with elongation and ellipticity of the sperm head. Sperms with larger head dimensions thus had larger MOT, PMOT, VCL, VSL, VAP, LIN, WOB, STR and smaller ALH and BCF. In contrast, sperms with greater elongation and ellipticity had lower motility and kinematic parameters. The effect of the extender (P < 0.05) and the storage time (P < 0.001) was highly significant for evaluated parameters. The ejaculates extended with INRA 96 reached significantly higher values (P < 0.01) for all evaluated parameters compared to Kenney extender. The INRA 96 extender significantly increased the values of PMOT, VCL and VAP compared to EquiPlus (P < 0.05). The results obtained from using INRA 96 showed that quality of cooled semen can be sufficiently maintained during at least 24 hours of storage.

The diploma thesis deals with the ram genital organs, hormonal regulation, sperm production and ejaculate. Practically were made preparations of rams before collecting semen, semen collection and ways of obtain semen. Further were evaluated quality native ram semen by macroscopically and microscopically methods. Evaluated material was obtained from zwartbles rams breed owned by Ing. Martin Hošek, Ph.D. All the ejaculate was immediately after collection examined macroscopically and microscopically. The volume of ejaculate, wave movement and activity of sperm were evaluated. Subsequently ejaculate transported was to the laboratory department of livestock reproduction of the Department of Animal Breeding Faculty of Agronomy, Mendel University in Brno. There was also assessed the concentration of sperm was made chill survival test when ejaculate was stored at 3 °C and the results were evaluated after 24 and 48 hours. There were also evaluated morphological coatings, which monitored defects of the head, acrosome defects, defects in the flagellum, immature sperm cells, sperm cells degeneration and normal sperm. The results show that the volume of the obtained sperm has an effect on genetic line, season, frequency of colection and age groups. For semen quality were proven decisive sampling sequence which had the effect of sperm concentration and survivability.

The aim of the study was influence of the complex of two elements (zinc, selenium) and two vitamins (vit C, vit E) on the organism of pigs under heat stress. Twenty boars of the Duroc breed were included in the experiment. The boars were approximately the same age and weight, housed individually. They were divided into 2 groups. The control group (n = 6) was fed only to a basic feed mixture containing 0.02 mg Se; 21.5 mg Zn; 9.9 mg of vitamin E and 16.0 mg of vitamin C. In addition, 0.5 mg of selenomethionine, 70 mg of alpha-tocopherol, 70 mg of zinc oxide and 350 mg of ascorbic acid were added to the diet (n = 6). The experiment took place from June to September at the insemination station in Velké Meziříčí. The control group showed a slight increase in ejaculate volume (by 19 %), which is common in the summer months. At the same time, the sperm concentration was unstable, it increased by 15 % in August, but fell below the baseline by 3.5 % in September. Motility was around 69 % during the experiment. Statistically significant was an increase (by 15 %) of morphologically abnormal sperm in the ejaculate (P < 0.05). The experimental group had higher ejaculate from the beginning. However, the volume curve was almost identical to the experimental group. The sperm concentration increased by 23 % in the trial group after adding antioxidants. During the experiment the concentration only slightly decreased. Motility in the experimental group averaged 70 % and did not change significantly during the experiment. Even in the control group there was a statistically significant increase in morphologically abnormal sperm (by 12.3 %), (P <0.05). From the results it can be concluded that the increased amount of selected antioxidants did not have a demonstrable effect on the improvement of the ejaculate quality, but it can be assumed that this quantity has ensured the stability of the seed parameters.

The aim of the thesis was to focus on the evaluation of ejaculate quality from rams obtained by various methods of sampling and description of ejaculate collection methods in male farm animals with a focus on small ruminants. The ejaculate collection method of rams in the zwartbles breed did not affect ejaculate parameters. A significant difference was found in the volume of ejaculate, and also in other indicators of ejaculate quality, for example in sperm activity, as well as in eddy motion, sperm concentration and percentage of morphologically normal spermatozoa. The most significant difference seems to be in sperm concentration; in other indicators, differences do not have such an impact on the production of insemination doses and practical utility in reproduction. The effect of the ejaculate collection method on rams was also evident in short-term preservation in commercial diluent and pasteurized milk.
